
Possible Incident Light Fluorescence Operational Problems
If normal adjustments are not getting the results you expect, check these:
• Weak fluorescence or image intensity: specimens improperly stored or too old
• Rapid fading of specimens i.e.: photobleached
• Unspecific or erroneous filter combination
• Numerical aperture of objectives too low
• Eyepiece magnification too high
• Failing or old mercury lamp
• Ambient light or room too bright
• Secondary light due to reflection at condenser
• Low contrast image due to excitation bandwidth too wide
• Unspecified staining
• Fluorescing mounting medium
• Auto-fluorescence of objective or immersion oil
• Dirty optics
